A scientist and his nephew are hosts of Lana, queen of the Amazons. Meanwhile, other white men penetrate the Brazilian jungle, wishing to find and take the legendary Amazons' treasure.
German Professor Van Vries (ANTON DIFFRING), and his nephew, Peter Van Vries (CHRISTIAN WOLFF), conduct a scientific exploration of the legendary El Dorado. The Professor is well learned on the natural dangers of the jungle, as well as of the curare venom used by the natives in their arrows. The expedition up the Amazonas, consists of the uncle and nephew team, and three Brazilian paddlers in a pirogue. They enjoy the natural beauty of the jungle, the large river basin, the high trees, the dense vegetation, the exquisite monkeys, the colorful birds, the birds of prey, the piranha fish eating a big animal afloat in the river.
They cross another pirogue, going down the river fast , from which a Priest in a white robe, shouts at them (in Portuguese) to be aware of dangerous white men ahead.
Indeed, Don Gerónimo de Araújo (DIETER EPPLER) is also heading to El Dorado, with a large number of servants and two local guides, a tall powerful black, Katango, and a shorter white Guide. Gerónimo punishes his servant Emmanuel severely, with a long bullwhip, and another man is so afraid of him that he tries to run away by , trampling on a number of alligators on the river shore. The man would have been eaten alive by one beast, if Peter had not shot it just in time, and rescued the wounded man.
By this chance encounter, the two groups meet, and Don Geronimo disarms Peter. The Van Vries are forced to join his group, as Gerónimo soon understood that the Professor would be helpful in taking him to the whereabouts of the treasure that he was after.
At the Great Falls, the adventurers are attacked by the Chavantes tribe, who make them all prisoners - except Peter who escapes wounded, and is taken care by a woman, who takes her to her tribe... of Amazons.
To Peter's amazement, their queen is a blonde young woman, Lana (MARION MICHAEL) who had been raised by the tribe as she was the only survivor of the airplane crash in the jungle. The natives considered her an envoy of the Heavens - their goddess. Peter is attracted to beautiful girl, and has plenty of occasions to see her topless or nude in the river bath - though her modesty is mostly covered by her long hairs, and the lion cub she frequently carries close to her bosom.
Peter convinces Lana to run away with him back to the civilized world. It is possible she is the long-lost granddaughter of a wealthy Hamburg industrialist... Tahira, Lana's handmaid, helps them to prepare their escape - knowing that the tribe will oppose it.
Meanwhile, Don Geronimo has made good in his plan, and discovered the cave where the gold and precious stones of the Amazons are. The honest Professor attempts to prevent Geronimo from touching a sacred stone, and his shot.
The Amazons give battle to Don Geronimo and his gang, and manage to kill all the invaders. The Professor dies in the arms of his nephew and Lana, and wishes them well.
Peter and Lana depart on a canoe down the Amazonas river, heading for their own adventure.
